چکیده

Gene regulatory networks are key elements for understanding how the different central nervous system regions differentiate. In recent years our group has done an extensive molecular characterization of the chicken pretectal region, obtaining a precise map of AP and DV molecular codes with respect to differential histogenetic pattern (Ferran et al., 2007; 2009, in press). As in other CNS regions, such as the spinal cord, Bmps signals from the roof plate and Shh signals from the floor and basal plates are clearcut candidates for governing the dorso-ventral patterning of the pretectum. The Msx1 transcription factor plays a role in diencephalic development, acting as intermediary between Bmp and Wnt (Bach et al., 2006), but it is still unclear how its expression is controlled by the gene regulatory network. Regulatory modules can be located in introns, exons or non-coding DNA regions, sometimes far away from the gene. In this work, we used comparative analysis of genomes to identify several highly-conserved non-coding DNA regions (HCNRs) located upstream and downstream of Msx1, and tested their ability for regulating Msx1 expression specifically in the pretectal region. This kind of approach is necessary for illuminating the specific regulatory network that patterns dorsoventral subdivisions in the pretectum, and possibly also in other CNS domains. Expression Analysis of RNA-Binding Motif Gene on Y Chromosome (RBMY) Protein Isoforms in Testis Tissue and a Testicular Germ Cell Cancer-Derived Cell Line (NT2)

a key factor in spermatogenesis and disorders associated with this protein have been recognized to be related to male infertility. Although it was suggested that this protein could have different functions during germ cell development, no studies have been conducted to uncover the mechanism of this potential function yet. Here, we analyzed the expression pattern of RBMY protein isoforms in test...

The histone genes cluster in Rhynchosciara americana and its transcription profile in salivary glands during larval development

In this work we report the characterization of the Rhynchosciara americana histone genes cluster nucleotide sequence. It spans 5,131 bp and contains the four core histones and the linker histone H1. Putative control elements were detected. We also determined the copy number of the tandem repeat unit through quantitative PCR, as well as the unequivocal chromosome location of this unique locus in...

Formation of Salivary Secretion in Rhynchosciara Americana

INTRODUCTION Since the genus Rhynchosciara belongs, like Chironomus, to the suborder Nematocera it will be very interesting to see if the formation of its salivary secretion proteins is similar to that of Chironomus secretion or to Bombyx silk biosynthesis . Moreover, this work will be of importance for a better understanding of the hemolymph protein function and will provide a basis for improv...

Gene Expression Profile Analysis during Mouse Tooth Development

Introduction: Complex molecular pathways involve in development of different tissues such as teeth. Differential gene expression patterns during teeth development generates different tooth types. Teeth development results from interactions between oral epithelium and underlying ectomesenchyme cells with neural crest origin. Teeth development are regulated by different signaling networks. In thi...

Ectopic Expression of Embryo/Cancer Sequence A (ECSA) in KYSE-30 Cell Line Using Retroviral System

Background Human preimplantation embryonic cells share many similarities with cancer cells such as ability to self-renew, unlimited proliferation and maintenance of the undifferentiated state. Embryo-cancer sequence A (ECSA), also known as developmental pluripotency associated-2 (DPPA2), is a cancer testis antigen (CTA) with unclear biological function yet. Objective: CTAs are expressed normal...
